To investigate the effects of traumatic stress and Astragalus Polysaccharide on cell immunity of mice amputee model. 50 BALB/C
mice were divided into 5 groups randomly, normal control group (group A), traumatic stress group (group B), traumatic stress+high dose APS group (group C), traumatic stress+middle dose APS group (group D), traumatic stress+low dose APS group (group E). CD 4,CD 8 and expression of c-fos protein were detected by immunohistochemisty method. Expression of NF-κB mRNA and IL-10 mRNA were detected by hybridization in situs.Their results were quantitated by HPIAS-1000 system. Compared with that of group A, the expression level of NF-κB mRNA and IL-10mRNA in thymus and spleen tissues of group B mice after injury 72 hours were elevated significantly(P<0.01),CD 4 and CD 4/CD 8 ratio were reduced(P< 0.01), the distribution of c-fos antigen more than that of group A.Compared with group B , the level of expression of NF-κB mRNA and IL-10 mRNA in thymus and spleen tissues were not only depressed by APS and the level of CD4 antigen, CD 4/CD 8 ratio were elevated in groups C,D,E,but thelevel of c-fos antigen were reduced(P< 0.01). Between the group B and the groupC,the expression of NF-κB mRNA and IL-10mRNA, CD 4 and CD 4/CD 8 ratio, the distribution of c-fos antigen had no significant difference (P>0.05). The cell immuni ty function of mice were disordered after their trauma,but APS may play a role in restoring their cell immunity.
