Objective:This subject is about immunoregulatory and treatment effect of LichangSiFang(LCSF)on molecular level by making
rat model of UC,especially they were observed the action of UC on
cytokine to make a progress for treatment UC with TCM.Methods:In the experiment,ninty-eight SD big rats were randomly divided into seven groups,Every group had fourteen rats(seven rats were male ,seven rats were female),namely the normal control group,the patholgic control group,the solfasalazine(SASP)group,the WuMeiWan(WMW)group,BaiTouWengTang(BTWT)group ,ShengllingBaiShuSan(SLSS)group,TongXieYaoFang(TXYF)group.Exceptthe normal control group,the other six animal model groups of UC were made through the integration of general immunity and local stimulation(DNCB and acetic acid).The six animal model groups of UC were treated by distilled water,SASP,TXYF,WMW,BTWT,SLBSS,TXYF respectively.After these treatment,the changes of some immune indexes in the colon tissue such as IL-6,IL-8,TNF-α,IL-10.Results:The level of IL-6?IL-8?TNF-α in pathologic group was significantly
higher than that in normal group(P<0.01),and the level of IL-10 was significantly lower than normal(P<0.01).The level of IL-6,IL-8,TNF-α in LiChangSiFan,SASP group was obviously lower than that in pathologic group(P<0.01),and the level of IL-10 was obviously higher than pathologic group(P<0.001),The level of IL-6?IL-8?TNF-α in WMW group,BTWT group,SLBSS group was lower than that in SASP group(P<0.05 or P<0.01),and the level of IL-10 was higher than SASP group(P<0.05 or P<0.01).The level of IL-6,IL-8,TNF-α in TXYF was higher than that in SASP group(P<0.05),and the level of IL-10 was lower than that in SASP group (P<0.05).The level of IL-6,IL-8,TNF-α in WMW group,BTWT group,SLBSS group is lower than that in TXYF group(P<0.05 or P<0.01),the level of IL-10 is higher than that in TXYF group(P<0.05 or P<0.01).The level IL-6,IL-8,TNF-αin WMW group and BTWT group was lower than that in SLBSS(p<0.05 or P<0.01),and the level of IL-10 was higher than that in SLBSS group(P<0.05 or P<0.01).The level of IL-6,IL-8 TNF-α in WMW group was lower than that in BTWT group(P<0.05 or P<0.01),and the level of IL-10 was higher than that in BTWT group(P<0.01).Conclusion:This model made with DNCB and acetic acid is successful.It is obvious that the cytokines are important role on UC especially proinflammtory cytokines IL-6,IL-8,FNF-α,which can promote inflammation,and immunosuppressive cytokine IL-10,which can restrain inflammation.LCSF could raise the index of IL-10 and decrease IL-6,IL-8,TNF-α(there are significant statistical different.)In the other word,they had curative effect by adjusting immunity and inhibiting inflammation reaction of abnormal enlarging and exacerbation in the course of UC.This implies us that we must have BianzhengLunzhi in treatment UC.
