RP-HPLC determination of insulin in WGA modified insulin liposomes

Objective:To develop a RP-HPLC method for determining the contents of insulin in
wheat germ agglutinin(WGA)modified
insulin liposomes and the entrapment efficiencies of the liposomes.
Methods:Insulin-liposomes were prepared by reversed-phase evaporation and sonication.Thh entrapment
efficiencies of the liposomes and the contents of insulin in the liposomes were analyzed by RP-HPLC.
Chromatographic conditions included Phenomenex-C_(18)(25 mm×4.6 mm,5 μm)and mobile phase consisting
of a mixture of acetonitrile-acetate trifluoride (0.01%).Detection wavelength was 210 nm.Results:The
calibration curve was linear in the range of 5~100μg/ml.The entrapment efficiencies of conventional li-
posome and WGA modified insulin liposomes were(30.30±2.77)%(n=3)and (69.33±4.54)%(n=3),respec-
tively.Conclusion:The method is convenient,accurate,reproducible,and suitable for determining the
contents of insulin in WGA modified insulin liposomes and the entrapment efficiencics of the liposomes.
