For the functioning of the transmission electron microscopy it is important that the specimens are very thin and they should be able to withstand in a high vacuum that occurs inside the instrument. From the transmission electron microscopy, it is possible to get the true 3D information with tilt-series-based tomography (Hoppe1981, Frank, 1992) or with serial-ultra-thin sections. (Sjostrand 1958, Stevens et.al, 1980). Among the many available techniques, one technique depends on choosing the accelerated voltage. Resolving the finest details for the specimens of special nature requires the usage of high voltage, an objective lens with low aberrations and a microscope that is highly stable in its electrical and mechanical aspects. Microscopes suitable for accelerating the voltages are very large and it requires mounting these microscopes in special suspension systems so as to isolate them from the site vibrations. There is a need to improve the lens design to reduce the image defects that arise from the spherical aberration. Using the electron velocity filters reduces the chromatic aberration. Lens asymmetry and selecting the focus are the factors that limit the resolution. The size of the stained molecules limits the resolution of the resolution of the stained structures. Better electron beam coherence improves the image contrast. To sort out the 3D structures, there exists an important technique, the stereo viewing technique. Very thin objects require high magnification and high tilt angle. Improving the resolution requires the acquisition of the speed of the microscopy which ultimately depends on the required signal-to-noise ratio ( RSN). The following formula estimates to achieve the minimally achievable voxel dwell time (τd) with back scattering coefficient (η).( Denk and Horsemann,2004) ( RSN )2e τd = ------------ ηIb where Ib is the beam of the current e is the elementary charge The available beam current depends on the electron-gun time, desired resolution and electron energy. Question: D Most of the drugs of the group that are immunomodulatory agents induce the lysosomal storage of the sulphonated glucosamino glycans (Fischeret.al,1996)There is a possibility for all the cells to develop abnormal cytoplasmic vacuoles that represent the ultra structural part of the lysosomal storage. The cell showing lysosomal accumulation of the glucoaminoglycans attributed to the impaired degradation. This is responsible for the functional changes as shown by the electron micrography. The flow cytometric technique shows the swelling of mitochondria that is the result of the opening of the permeability transition pores of the mitchondrial membrane eventually resulting in the collapse of the transmembrane potential of the mitochondria and the release of the cytochrome c into the cytosol from the mitochondria. The pathological conditions that result from the swelling of the mitochondria include swelling and the formation of the megamitochondria. This hypothesizes that this has a direct relationship with the apotopic processes of the cell.
