Determination of Bis (2-ethylhexyl) phthalate in water by Gas Chromatography with Micro Electron Capture Detector (GC-μECD) and confirmation by Gas Chromatography with Mass Spectrometry (GCMS)by Azharuddin Abd Aziz, Norhaida Abd. Latiff, Mohan Gangadharan
Phthalates, or phthalate esters, are a group of chemical compounds that are mainly used as plasticizers (substances added to plastics to increase their flexibility). There are different types of phthalates, each varying in chemical structure and molecular weight. The physical forms of these compounds are liquids similar in appearance and consistency to vegetable oils.1,2 A study conducted on rodents involving intake of large amounts of Phthalates has shown that phthalates cause damage to the liver, kidneys, lungs and developing testes. In 2005, phthalates were reported to mimic the female hormone estrogen and cause "feminisation" of baby boys.2 The National Standard for Drinking Water Quality (NSDWQ) stipulates a maximum acceptable level (MAL) of 8ppb in drinking water for Bis (2-ethylhexyl) phthalate which is listed as a potential carcinogen in the National Toxicology Program, USA. There is no specified level for other phthalates in the National Standard. A gas chromatography micro electron capture detector method has been reviewed to determine the concentration of Bis (2-ethylhexyl) phthalate in drinking water. This method is based on the USEPA Method 8061A3 (for aqueous and solid sample by GC-ECD) and 5064 (for drinking water by GCPhotoionization). Instrumentation part was adapted from method 8061A whereas extraction part was adapted from method 506. A parallel, dual column GC-μECD was used for the determination, with the primary column DB-1701 column used for quantitation while the secondary column DB-5 used for confirmation. Confirmation was also was carried out using GCMS. The structure of this compound is shown in Figure 1. Recovery and validation studies were carried out for Bis (2-ethylhexyl) phthalate at the level of compliance with the NSDWQ.
