Objective It is to observe the influence of
Poge saving heart decoction(PSHD) on myocardial cell's apoptosis of
heart failure
rat after myocardial infarction.Methods The
heart failure models were made with 36 Wistar rats and randomly divided into PSHD group(12 rats),Captopril group(12 rats) and model group(12 rats).Other 12 Wistar rats not operated on were chosen as control group.PSHD group were given PSHD 10 mL/kg twice a day by intragastric administration.Captopril group were given Captopril liquid 10 mL/kg twice a day by intragastric administration.Model group and control group were given drinking water 10 mL/kg twice a day by intragastric administration.Body mass was weighed once a week to adjust the dosage of drugs.In the third and sixth test week,the rat hearts were separately made into paraffin slices.Cardiac muscle cell apoptosis was examined with TUNEL method.The expressions of apoptosis regulative genes bax and bcl-2 protein were detected with immunohistochemical method.Results The positive apoptosis cells in the brim area of myocardial infarction in PSHD group and Captopril group reduced much more than those in model group,the apoptosis rate was lower especially six weeks after administer.The average light intensity of bax protein expression remarkably increased and that of Bcl-2 protein expression obviously decreased in PSHD group.Conclusion PSHD can improve the apoptosis of heart failure myocardial cells after cardiac infarction,which may be related with its changing the cell apoptosis regulative genes.
