Objective: To establish a method for the determination of the contents of four
active ingredients baica-lin,
wogonoside,
baicalein and
wogonin in
Radix Scutellariae of different resource and to study the difference of chemical
constituents in them by gradient elution HPLC. Methods:34 samples of Radix Scutellariae from different habitat, specification or species were collected, four active constituents in them were determined by RP - HPLC method. Meanwhile, the data of whole chemical constituents of 34 samples were obtained from the chromatograms and analyzed by Hierarchical Clustering method. The
column used was Platinum ODS(250 mm×4. 6 mm) with 5 μm packing, the mobile phase used were A : acetonitrile - water - formic acid( 17: 83: 1 ) ; B : acetonitrile - water -formic acid(80: 20: 1 ) , with gradient elution,the column temperature was controlled at 40℃ ,the detection wavelength was set at 280 nm. Results:This method is accurate and reproducible as well as specific. As the result of Hierarchical Clustering, S. baicalensis ( different resources or prepared pieces ) and the other species of Radix Scutellar-iae are classified to different sort. Conclusion:The contents of the four active constituents in S. viscidula,S. rehderi-ana or S. amoena are similar to that of Radix Scutellariae, but the whole chemical constituents is considerably different from Radix Scutellariae.
