Objective To investigate the effects of
ginsenosides, Rg1 and Rb1 on ex vivo amplification of CD34 + cell isolated from
cord blood. Methods The cells were amplified in StemSpanTM SFEM serum-free medium which was added to GS, Rg1, Rb1 of different concentrations for counting total cell numbers and monitoring
Cd34 +Cell. After amplification, the cells were cultured in semisolid medium of MethoCult TM H 4435 in order to count CFU, BFU-E and CFU-GM colonies. Results GS, Rg1, Rb1 could stimulate CD34 +cells in liquid culture system at the best concentrations of 50 μg/ml, 5 mg/ml and 5 mg/ml. The numbers of total cells in GS, Rg1, Rb1 groups were amplified by 20.4, 24.5 and 24.1 while the CD34 +cell numbers were amplified by 4.22, 5.19 and 4.12. After cultured in liquid, CD34 +cell could still stimulate the formation of colony of amplified cells. The abilities of the formation of colony of CFU-E were 90.1%, 70.8% and 72.8% higher than that in control group, the abilities of the formation of colony of BFU-E were 53.4%, 68.0% and 67.0% higher than that in control group, and the ability of the formation of colony of CFU-GM were 67.0%, 29.8% and 39.4% higher than that in control group. Conclusion GS, Rg1 and Rb1 can stimulate proliferation and orienting differentiation of the hematopoietic stem cell.
