Objective\ Hematopoietic growth factors (HGF) play a very important role in hematopoietic regulation. Saponins is known as
of the main effective fractions in Chinese traditional medicine Panax Ginseng. Our previous study has showed that total saponins of panax ginseng(TSPG) could stimulate hematopoiesis significantly. The present study investigate the effect of TSPG on the bioactivity and mRNA expression of HGF for clarifying the hematonic mechanism of Panax Ginseng. Methods\ The techniques of nucleic acid
in situ hybridization, culture of hematopoietic progenitor cells ex vivo and bioassay of HGF were used. Results\ TSPG could markedly enhance bone marrow stromal cell (BMSC), peritoneal macrophage(PMφ) and splenic cell (SPC) to express GM CSF mRNA and IL 6 mRNA. The BMSC, PMφ and SPC conditioned media, prepared by treating these cell with TSPG (20mg/L) ex vitro , could obviously promote proliferation and differentiation of BFU E, CFU GM and CFU MK. These mean that much higher burst promoting activity (BPA), granulocyte and macrophage colony\|stimulating activity (GM CSA) and megakaryocyte colony\|stimulating activity (MK CSF) exist in TSPG induced conditioned media. Conclusion TSPG may regulate hematopoiesis by activating GM CSF and IL 6 mRNA expression of BMSC. Mφ and lymphocyte in hematopoietic inductive microenvironment. This mayht be one of the mechanisms for the promoting vital energy and hematopoiesis of panax ginseng.\;
