AIM To study the effects of
prostaglandin E 1 (PGE 1) and ligustrazine(LT), alone or in combination, on reperfusion arrhythmias
in anesthetized rats. METHODS The arrhythmias were produced by coronary artery ligation for 30 min followed by reperfusion 60 min. Superoxide dismutase(SOD) and
glutathione peroxidase (GSH Px) activities, malondialdehyde(MDA) and Ca 2+ contents in myocardium, serum creatine Kinase MB(CK MB) were measured by thiobarbituric acid fluorescence assay, colorimetric determination and atomic absorption photometry. RESULTS PGE 1 (31 15 μg·kg -1 ) in combination with LT (25 mg·kg -1 ) were more effective on prevention of the reperfusion arrhythmias than each drug alone at more dose. The combination of both drugs not only decreased the contents of MDA, Ca 2+ and CK MB, but also increased the activity of SOD and GSH Px in reperfusion myocardium. CONCLUSION PGE 1 in combination with LT in half of individual dose has protective effects on myocardial ischemia reperfusion injury and there was some synergic effect when they were used in combination. The protective effect may be from that they can increase the activity of clearence enzyme of free radical, inhabit lipid peroxidation, and the calcium influx.
