AIM:To observe the protective effect of polarized liquid (insulin) on
myocardial ischemia reperfusion injury in rabbits.
METHODS:Forty eight healthy male rabbits were randomly divided into 3 groups of 16 each:control group,insulin group and glucose insulin potassium(GIK) group.Rabbit models of
myocardial ischemia reperfusion(MI/R) were established,and were treated with GIK,insulin and saline respectively.The size of myocardial infarction was detected after reperfusion,and ultrastructure of myocardial cells was examined under electron microscope.
Apoptosis was identified by TUNEL and the contents of protooncogene(Bcl 2) and Fas were measured by immunohistochemical technique. RESULTS:The injuries of ultrastucture in the GIK and insulin group were lighter than that in the control group,and the sizes of myocardial infarction were reduced obviously
(q=4.32, 3.95,P< 0.05). Apoptosis indexes in the GIK group(1.38±0.82) and insulin group(1.28±0.54) decreased markedly compared with that in the control group(2.15±0.19)(q=0.65, 0.72,P< 0.05),so was the Fas contents (q=0.07, 0.06,P< 0.05),while the Bcl 2 contents increased in the GIK group and insulin group compared with the control group(q=0.14,0.11,P< 0.05).There were no significant differences in apoptosis index,contents of Fas and Bcl 2 protein between GIK and insulin groups(P >0.05).GIK(insulin) can protect myocardium from ischemia reperfusion injury by modulating the expressions of Fas and Bcl 2 protein and inhibiting apoptosis after myocardial ischemia reperfusion.
