Objectives>To investigate the effect of Cordyceps polysaccharide
liposomes (CPL) on the
expression of transforming factorβ 1
messenger RNA (TGF-β 1 mRNA) in hepatic tissues and its inhibitive function on the development of immune hepatic fibrosis (IHF) induced by concanavalin A(Con A) in BALB/c mice.One hundred female BALB/c
mice were divided into 4 groups:①Group of normal control (n=10);②Group of positive control , treated with Con A only(n=30); ③Group treated by small dose of CPL(10mg/kg, qd. ×20 wk) plus Con A (n=30) and ④Group treated by large dose of CPL(30mg/kg, qd. ×20 wk) plus Con A(n=30). Mice models of IHF were established by intravenous injections of Con A weekly (via the retro-orbital venous plexus under anesthesia). Normal saline was administered in mice of control group. Blood samples were collected by venous puncture from mice at the 12th
wk after Con A or CPL administration and serum activities of alanine aminotransferase (ALT), aspartic acid aminotransferase (AST), serum levels of albumin, hyaluronic acid (HA), procollagen Ⅲ(PC Ⅲ) and collagen Ⅳ (IVC) were estimated. Total RNAs were extracted from liver tissues and TGF-β 1 mRNAs were analyzed by one step reverse transcriptase-polymerase chain reaction(RT-PCR) in 5th,12th and 20th wk. Each 10 mice in ②,③ and ④
groups were decapitated at these same time intervals. Using b-actin as the internal marker, the relative expression levels of TGF-β 1 mRNA were calculated. The liver function and extent of hepatic fibrosis in all groups were observed in 12th wk during experiment.Serum levels of ALT, AST, HA,PC Ⅲ and Ⅳ C in CPL-treated groups were obviously lower than those in Con A group ; while the relative expression levels of TGF-β 1 mRNA in CPL-treated group were dosage-dependent and also significantly decreased as compared to those in Con A group.
CPL may inhibit the occurrence of IHF in mice induced by Con A through down-regulation of TGF-β 1 mRNA expression.
