AIM:To evaluate pharmacokinetic characteristics of isosorbide 5 mononitrate(5 ISMN) tablet, 5 ISMN injection, and isosorbide dinitrate(ISDN) injection in Chinese healthy volunteers. METHODS: Eighteen healthy male volunteers received a single dose of 20 mg ISDN intravenous injection, 20 mg 5 ISMN tablet or 20 mg 5 ISMN intravenous injection in self cross administration. Each plasma concentrations of ISDN and 5 ISMN were measured by a validated gas chromatographic method. RESULTS: The main pharmacokinetic parameters of 18 volunteers after oral administration of the 5 ISMN tablet and intravenous drip of 5 ISMN injection were T max (0.7± s 0.6), (4.1± 0.7) h; C max (389±86),(283±59) μg· L -1 ;AUC 0 ∞ (3 266±624), (3 179±769) μg·h·L -1 . The main pharmacokinetic parameters of ISDN,2 ISMN and 5 ISMN were C max (18±7),(59±4),(209±50)μg· L -1 ,respectively; AUC 0 t (85±28), (380±59), (1 384± 427)μg·h · L -1 , respectively; T 1/2 (1.00±0.20),(2.6±0.8),(6.1±0.9)h, respectively after intravenous drip of ISDN. CONCLUSION: 5 ISMN tablet compares with 5 ISMN injection, the AUC of them is equivalent. But the former is faster than the later in onset time. ISDN injection can shortly reach stable concentration.
