AIM:To study protective effects of the taurine on cisplatin-induced damage in primary cultures of kidney proximal tubular
cells(PTC).Meth-ods:Primary culture of rabbit proximal tubular cells was established.Ethidium Bromide binding assayed DNA interstrand crosslink and cytosolic free 〔Ca 2+〕 i was measured with Fura-2/AM. In cisplatin group,PTC was incubated without taurine and cisplatin for 24 hours, and with cisplatin (26μmo1.L 1)for another 24 hours.In taurine group,PTC preincubated with taurine(0.1,1,10g.L 1)for another 24 hours,and cisplatin(26μmo1.L -1 was added into culture for another 24 hours. In control group, PTC was incubated without cisplatin and taurine for 48 hours. Results:(1)After culture with cisplatin for 24 hours, PTC was induced DNA interstrand crosslink and cytosolic free 〔Ca 2 +〕i overload,preinculture with taurin (1, 10g.L 1)before 24 hours, these effects on PTC were provented. Conclussion: Taurine could protect cisplatin-induced damage of PTC.
