OBJECTIVE To study the protective effect and mechanism of total flavone extract of Elsholtzia blanda (TFEEB) on acute myocardial ischemia. METHOD Rats were given TFEEB for 5 d by ig . Rat acute myocardial ischemia models were prepared by iv pituitrinum(pit)in the 5th day. Degree of ischmia was assessed by T amplitude of lead Ⅱ dynamic ECG and heart rate. The mechanism of anti-myocardial ischemia was investigated by the level of creative phosphokinase(CK), calcium(Ca 2+), nitric oxide(NO) in serum and Ca 2+, NO, malondialdehyde(MDA)in myocardial tissue. RESULTS At the dose of 200 mg/kg, TFEEB could markedly reduce the occurrence of arrhythmia, reverse the elevated T amplitude of ECG and slow heart rate induced by pit(P<0.05). The serum level of CK and myocardial Ca 2+, MDA were decreased and Ca 2+in serum, the level of NO in serum and myocardial tissue were significantly increased in the TFEEB group(P<0.05 or P<0.01). CONCLUSION The results suggest that the protective effect of TFEEB to myocardial ischemia in rats is related to prevent calcium overload, increase the level of NO in myocardial tissue and inhibit lipid peroxidation.
