The velvet antler polypeptides were extracted with acid and alcohol,which showed some bands below 17kDa in SDS-PAGE and a
single band in Western-boltting with the anti-human IGF-1 as the first antibody.Quantitative analysis showed that the polypeptides was 1~2 mg/g of green weight by Bradford technique,IGF-1 was 300~400 pg/g by radioimmunoassay(RIA).Mice were injected with polypeptides 1,4,8 mg/kg of body weight,after six days,the experiments on growth promotion, immunological regulation,anti-lipid peroxidation,anti-fatigue were done.The results showed that:The mean accumulative weight gained in polypeptide group was higher than that of the control group,but not significantly(P>0.05);The rosettes looping rates of the 1,4,8 mg/kg polypeptide groups were 33.3%,34.2%,39.6% higher than the control(injected with NS)(P<0.05);OD value of haemolysin in serum of 8 mg/kg polypeptide group was significantly different from the control(P<0.01);The phagocytosis percentage of the 1,4,8 mg/kg polypeptide groups were 22.1%,25.8%,26.5% higher than the control(P<0.05,(P<)0.01),which indicated that the polypeptide has the function of immunological regulation.The superoxide dismutase(SOD) activities and malondialdehyde(MDA) content in serum of old mice injected with 4,8 mg/kg polypeptide groups 51.1%,59.2% higher and 14.9%,16.7% lower respectively than the control group(P<0.05),which indicated the anti-lipid peroxidation function of the polypeptides.In the weight swimming experiment,the swimming time of 8 mg/kg polypeptide group was(21.67 mins) longer than the control(P<0.05),which indicated the anti-fatigue function of polypeptides.
