Objective To review the pharmacokinetic behavior of cefoperazone(CFP)and tazobatan(TAZ)in compound preparation(CFP:TAZ=8:1)in
dogs and their mutual influences.Methods To 4 Beagle dogs,single i.v.injection of CFP(110 mg/kg)、TAZ(56 mg/kg)and compound preparations with 3 different dosages(31、125 and 500mg/kg)separately.The concentrations of CFP and TAZ in serum were determined by RP-HPLC,the interpolated pharmacokinetic curve was drawn and the main pharmacokinetic parameters were calculated.Results The dispositions fitted two compartment model in Beagle dog,after single i.v.injection of CFP、TAZ and compound preparation with CFP and TAZ;Analysis of variance appeared to be no difference after single injection of CFP(110 mg/kg)and equal dose CFP in compound preparations.The mean pharmacokinetic parameters(T1/2α、T1/2β、 Vc and CLs)appeared to be no difference after single injection of CFP and 3 different dosage of compound preparations.In vivo pharmacokinetic appeared to be linearity.The dispositions fitted two compartment model in Beagle dog,after single i.v.injection of TAZ(56 mg/kg)and compound preparation(500 mg/kg including TAZ 55.6 mg/kg).There was no obvious difference by comparison of the concentration in serum and pharmacokinetic parameters.Conclusion Single injection of CFP、TAZ and compound preparation with CFP and TAZ to Beagle dogs appears to be no mutual influence in pharmacokinetic studies.It implies that it is theoretically feasible to develop new preparation with CFP and TAZ.
