Objective To evaluate the effect of
Tripterygium wilfordii on the expression of Transforming Growth Factor(TGF)-β_1 in
bronchial epithelial cells stimulated by Interleukin(IL)-1β and to investigate the molecular mechanism of protective effect of Tripterygium wilfordii(TW) on
airway remodeling. Methods The normal cultured bronchial epithelial cells were divided randomly into 5 groups:normal control group(incubated with equal serum-free conditioned medium),IL-1β treatment group(exposed to 10 ng/ml of IL-1β for 24 h)),both IL-1β and TW treatment groups A, B and C which were stimulated by IL-1β 10 ng/ml for 24 h after 30 min in the presence of TW(5 μg/ml, 10 μg/ml, 20 μg/ml , respectively), then the expression of TGF-β1 was measured by immunocyte-chemistry. Results The expression of TGF-β_1 in IL-1β treatment group(0.511±0.012)were significantly higher than that in control group (0.138±0.009,P<0.01). The expression of TGF-β_1 in IL-1β together with TW treatment groups A(0.353±0.046), B(0.245±0.034), and C(0.218±0.024)were significantly lower than that in IL-1β treatment group (P<0.01). TW treatment caused dose-dependent inhibition of IL-1β-stimulated expression of TGF-β_1. TW in a dose range of 5-20 μg/ml showed effectively inhibition to TGF-β_1 . Conclusion TW can remarkably decrease IL-1β-induced over-expression of TGF-β_1 in bronchial epithelial cells. The ability of IL-1β to modulate TGF-β_1 release may contribute to both abonormal airway repair and development of airway remodeling. TW can suppress the development of
airway remodeling by downregulating the over-expression of TGF-β_1 stimulated by IL-1β, so it can be inferred that it is one of molecular mechanisms of the protective effect of TW on airway remodeling.
