AIM: To observe the protective effect of
naoyikang serum on PC12 cell injury induced by glutamate(Glu) and
hydroperoxide with serum pharmacology method.METHODS: ①The experiment was conducted in the Laboratory of Pathophysiology, School of Basic Medical Sciences, Nantong University from October to December 2004.24 SD rats aged 12 months were randomly divided into 2 groups:
Control group and naoyikang gastric perfusion group (naoyikang group) with 12 rats in each group. Rats in the naoyikang group received gastric perfusion of naoyikang at 1.5 times of equivalent dose for 10 continuous days, serum after taking naoyikang was collected one hour after the last gastric perfusion. Serum in the control group after gastric perfusion of normal saline of the same volume was collected as control. ② Seven days after induced by nerve growth factor (NGF) of PC12 cell, it was flushed twice with DMEM basic medium, which was added 100, 300 and 500 g/L serum of naoyikang respectively, incubated at 37 ℃ for one hour, then incubated by added 0.2 mmol/L
glu and 0.5 mmol/L hydroperoxide for another 30 mins. Meanwhile,
model group (PC12 cell injury model induced by Glu and hydroperoxide without adding Glu and hydroperoxide) and naoyikang group (rat's serum without added Glu and hydroperoxide) were set up.③Survival
rate of PC12 were assayed with MTT colorimetry; Releasing rate of lactic dehydrogenase (LDH), content of malondialdehyde (MDA) and superoxide dismutase (SOD) activity were assayed according to the instruction on the test kit.④Differences of measurement data were compared with ANOVA method. RESULTS: ①Survival rates of PC12 cell in the Glu group and hydroperoxide group were significantly lower than that in the control group(P < 0.05,0.01); While releasing rate of LDH were obviously higher than that in the control group(P < 0.01);The content of MDA of PC12 cell in the Glu group and hydroperoxide group was obviously higher than that in the control group (P < 0.01); While SOD activity was significantly lower than that in the control group (P < 0.01).②Survival rate of PC12 cell in the naoyikang group was obviously higher than that in the Glu model group and hydroperoxide model group (P < 0.05-0.01); While the releasing rate of LDH was significantly lower than that in the Glu model group and hydroperoxide model group (P< 0.05-0.01).③Content of MDA in the naoyikang group was significantly lower than that in the Glu model group and hydroperoxide model group(P < 0.05,0.01); While SOD activity was significantly higher than that in the Glu model group and hydroperoxide model group(P < 0.05,0.01=.CONCLUSION: Naoyikang serum can ameliorate the PC12 cell injury induced by Glu and hydroperoxide, its mechanism may relate with the decreasing of generation membrane lipid peroxide and the increasing of activity of antioxidant enzyme.
