AIM: To investigate the early interventional effect of xingding
solution extracted from ginkgo leaf on ototoxic deafness
of guinea pigs induced by
gentamycin. METHODS: ① The experiment was completed at Chuannan Audile and Language Disease Prevention and Cure Center Affiliated to Luzhou Medical College from November 11, 2003 to May 1, 2004. Totally 24 guinea pigs with normal distortion product otoacoustic emission (DPOAE) were randomly divided into 3 groups: gentamycin model group (n=8, 16 ears), gentamycin+ xingding
solution group (n=8, 16 ears) and control group (n =8, 15 ears). ② Guinea pigs in gentamycin model group were injected with 100 mg/kg gentamycin (Southwest Pharmacological Limited Company) for 7 successive days. Guinea pigs in gentamycin+ xingding solution group were injected with 100 mg/kg gentamycin+0.4 mL/kg xingding solution (Guizhou Yibai Pharmacological Limited Company) for 7 successive days. Guinea pigs in control group were injected with 0.9% saline for 7 days. ③ DPOAE was monitored to observe the amplitude, and analyzed the change of amplitude of DPOAE at the frequencies of 1, 2, 4, 6 kHz among 3 groups 3 and 7 days after injection. ④ Measurement data were compared with t test. RESULTS: Totally 24 guinea pigs with 47 ears entered the final analysis. ① Amplitudes of DPOAE at 4 and 6 kHz in gentamycin model group were higher than those in gentamycin+ xingding solution group 3 days after injection (23.19±3.80), (26.40±4.70) dB SPL; (24.88±5.12), (29.96±5.74) dB SPL, t=2.079, 3.763, P < 0.05; but difference at 1 and 2 kHz between two groups was not significant (P > 0.05). Differences of amplitude at 4 frequencies between gentamycin+ xingding solution group and control group were not significant (P < 0.05). ② Amplitudes of DPOAE at 4 and 6 kHz in gentamycin model group were lower than those in gentamycin+ xingding solution group 7 days after injection (21.39±3.04), (24.83±3.27) dB SPL; (23.90±4.35), (27.90±4.15) dB SPL, t=2.239, 4.269, P < 0.05; but difference at 1 and 2 kHz between two groups was not significant (P > 0.05). Amplitudes at 2, 4 and 6 kHz were lower in gentamycin+ xingding solution group than those in control group 7 days after injection (t=3.127, 2.121, 2.352, P < 0.05). CONCLUSION: Xingding can inhibit damage of gentamycin from cochlear hair cell during early period.
