Objective To observe the effect of genistein(Gen) on
testosterone level in
serum of benign
prostatic hyperplasia(BPH)
mice induced by testosterone propionate.Methods BPH model mice were induced by subcutaneously testosterone propionate to detect prostatic
wet weight and to calculate prostatic wet weight index(PI).The morphological changes of prostate were observed through light
microscope and electron microscope.Serum androgen level was determined by radioimmunoassay.The glandular morphologic parameters of prostate tissue were measured with image-analysis.Results The prostate wet weight index
decreased dosedependently 1 week after treatment with Gen(20,40,160 mg/kg) for three weeks. Under light microscope,the enlarged epithelia and papillary configuration decreased or disappeared,noted cubical or flat epithelium,expanded or shrinked gland cavity.The image-analysis showed that mean maximum and minimum diameter and mean glandular area of prostate decreased(P<0.05).Through electron microscope,noted the regularly lined epithelia with nuclear lied at the base of cells,decreased secretory granules,developed Golgi complex,the dropped microvillus and nucleus into the glandular lumen.Conclusions Gen could obviously inhibit BPH and decrease serum testosterone level,which suggests that the inhibition mechanism of Gen on BPH might be involved in anti-androgen.
