OBJECTIVE To study the intervention by extract from
Cuscuta chinensis in the structure of human sperm membrane and the oxidative
injury of function from active oxygen(ROS) and the protection of the function of human sperm membrane.And to study the mechanism about
Cuscuta chinensis treating male's infertility and asthenoospermia.METHODS To select sperm with normal physiological function using the Percoll gradient centrifugation for the normal sperm model,and separate the sperm suspension into the normal group,the model group and the control group(Vitamin C group),and the massive,the medium and the small dose of Cuscuta chinensis.The ROS was made from hypoxanthine-xanthine oxidase(HX-XO),and ROS,different content(0.125,0.25,0.5 g·L~(-1)) of extract were hatched with sperm in the oxygen environment,the sperm membrane lipid peroxide injury were analyzed,and the function of sperm membrane were analyzed by sperm hypoosmoticswelling(HOS).RESULTS In the same conditions,the small,medium and massive extract from Cuscuta chinensis(0.125,0.25,0.5 g·L~(-1))improved SOD vitality of sperm suspension,reduced the content of MDA,intervened the injury of sperm membrane by ROS to some extent and protected some function of sperm membrane.The 0.125 and 0.5 g·L~(-1) extract had no obvious difference(P>0.05) with Vitamin C(0.25 g·L~(-1)) in it,but the content(0.25 g·L~(-1)) of extract is significantly better than control VitC(P<0.001).CONCLUSION The suitable content of extract from Cuscuta chinensis can significantly improve the SOD vitality of sperm suspension,reduce the content of MDA,and intervene
lipin peroxidation by ROS in sperm membrane by the way against oxidation,significantly protect the structure and function of sperm membrane,that is one of the mechanism for Cuscuta chinensis treating male's infertility and asthenoospermia.
