AIM To compare the
analgesic effects of tetrodotoxin TTX alone acetylsalicylic
acid ASA alone and
combination of TTX and ASA. METHODS The experiment was conducted at the Department of Pharmacology School of Basic Medical Sciences Peking University between October 2003 and March 2005.Totally 230 healthy KunMing
mice were randomly divided into TTX group ASA group and combination group including 23 subgroups with 10 in each subgroup. The reinforced analgesia and synergistic effects of the combination were examined by using the acetic acid-induced writhing test in mice. RESULTS Totally 230 mice were involved in the result analysis.①TTX group When the mice were injected with 0.5-4.0×10-3 mg/kg TTX the inhibitory dose ID50 of mice was 2.1×10-3 mg/kg When 4.0×10-3 mg/kg TTX was used the inhibitory rate on acetic acid-induced writhing response of mice was 65.2%. ②ASA group When injected with 12.5-200×103 mg/kg ASA the ID50 was 64.0 mg/kg and the corresponding rates of analgesia inhibition were from 15.8% to 93.2% respectively.③The analgesic effects were obviously increased at the small dose of TTX 0.79×10-3 mg/kg 0.39×10-3 mg/kg plus ASA. The ID50s of ASA were reduced to 12.6 mg/kg and 5.8 mg/kg respectively. CONCLUSION The small dose of TTX combined with ASA can markedly increase the analgesic effects.
