Objective To observe the effect of
tetramethylpyrazine (TMP) on the hemorheology in
pressure overload-induced hypertensive rats.Methods Twenty-two SD
rats were randomly divided into operation (OG), operation plus TMP low concentration injection (TMP 1), operation plus TMP high concentration injection (TMP 2) and sham operation (Control) groups. The pressure
overload hypertensive models were established by contracting abdominal aorta of rats and drinking 1 % NaCl.After three weeks operation,ligustrazine hydrochloride injection were administed (40mg/kg/d, TMP1 group; 80mg/kg/d,TMP2 group) by intraperitoneal injection.Two weeks later, the arterial blood pressure, electrocardiogram and hemorheological indexes were measured. Results The mean arterial blood pressure in OG or TMP1 or TMP2 groups was
higher than those in control group, P<0.01. Under shear rate 10,60,120s-1,whole blood viscosity or whole blood reduced viscosity in OG group were higher than those in control group, P<0.05 or P<0.01. Hematocrit, aggregation index of erythrocyte and rigidity index of erythrocyte in OG group were significantly higher than those in control group, P<0.01, compared with control group. In TMP 1 group,these responces of hemorheology were similar to those in OG group. And in TMP 2 group, above mentioned indexes were depressed compared with OG group, near those in control group. Conclusions Pressure overload could let to
hypertension and increase of blood viscosity, hematocrit, aggregation and rigidity of erythrocyte.These changes can be improved by TMP of higher concentration.
