AIM:To observe the therapeutic effect of
guizhi fuling capsule on hyperplasia of
mammary glands in experimental
rats model and its mechanism. METHODS:SD rats were randomly divided into control group,model group,guizhi fu ling high
dosage group,middle dosage group and low dosage group according to the ir body mass.Rats model of hyperplasia of mammary glands were reproduced by inje cting extrinsic estradiol(E2) and progesterone(Pt) into muscle.Morphologic chang es of nipple were observed in every group.The content of
serum E2,Pt,interleukin 2 (IL 2) and interleukin a(TNF a) were measured with radioimmunoassay metho d.The changes of estrogen receptor(ER) and androgen receptor(PR) were observed w ith immunohistochemical method. RESULTS:In the guizhi fuling capsule treatment group,the hyperplastic coats of duct in mammary glands,the number of acinus in lobule,the content of serum TNF a and E2 in plasma,and the score of ER and PR in mammary glands tissue were si gnificantly decreased.The content of serum
pt and IL 2 in plasma was significan tly increased.Compared with the model group,there was significant difference(t=2 .31-3.16,P< 0.05 or t=3.45-13.21, P< 0.01).The content of serum E2<(6.25±3.18 ) ng/L>in the high dosage group was significantly decreased than that in the mod el group<(25.22±11.45) ng/L>(t=5.05, P< 0.01); the content of Pt in the high do sage group<(3.68±1.33) μg/L> were significantly increased than that in the mod el group<(2.11±0.84) μg/L>(t=3.16, P< 0 .05). CONCLUSION:Therapeutic action mechanism of guizhi fuling capsule on hyperplasi a of mammary glands in rats may relate with adjusting the endocrine and immune f unctions of experimental rats.
