To investigate the effect of Oxymatrine (OMT) on mouse lymphocyte proliferation stimulated by Con A through fluorescence dyestuff CFDA-SE. Utilized CFDA-SE dyed and then flow cytometer to examine the fluorescence intensity changes of lymphocytes co-influenced by polyclonal stimulator Con A and OMT. Whereafter, analyze the degree of effect of OMT on mouse lymphocyte proliferation by related software. After culture 48 h, CFSE fluorescence could be examined by cytometer, while control group did not appear sub-peak . It was also to say that lymphocytes did not proliferate. Three peaks were obvious in Con A group. This means that Lymphocytes appeared cleavage after 48 h stimulated by Con A. Their exhibition was the fluorescence density of CFSE halved. In all groups of Con A+OMT treated, high does (OMT500 μg/mL) groups , Aboriginal peaks all higher but sub-peaks lower than those of groups of Con A significantly. However, it was no significant difference between aboriginal peaks and those of control groups. This means OMT obviously restrain lymphocyte proliferation. The results of middling dose (OMT 125 μg/mL) groups and those of high dose groups were consistent. This showed that middling dose of OMT could also obviously restrain mouse lymphocyte proliferation.
The results between low dose (OMT 31 μg/mL) groups and those of Con A groups had no significant differences. But compared to control group, the fluorescence intensities of results of all groups had significant differences. It showed that OMT of this dose couldn't significantly restrain lymphocyte proliferation. After culture 72 h, the fluorescence intensity changes between all groups consistent to those of culture 48 h . Though CELLQuest software analyzed, the educed proliferation guidelines showed that OMT respectively in 500 μg/mL, 125 μg/mL and 31 μg/mL could restrained the promote effect of Con A on lymphocyte proliferation in manner of dose dependence. CFDA-SE dyeing and flow cytometer were both reliable tools to analyze lymphocyte proliferation. OMT respectively in 500 μg/mL, 125 μg/mL and 31μg/mL can restrained the promote effect of Con A on lymphocyte proliferation in manner of dose dependence. Namely, OMT can obviously restrain mouse lymphocyte proliferation.
