AIM:To investigate the therapeutic effect of
guishen capsule on
serum concentr ation of estradio(E2),progesterone(P) and prolactin(PRL) in rat models of mammar y
gland hyperplasia. METHODS:The experiment was completed in the Function Laboratory of College of Medical Sciences,China Three Gorges University from May to July 2004. Forty eig ht unfertilized female SD rats in clean grade weighing (200±20) g were randomly divided into 6 groups with 8 rats each. ①Rats in the control group were fed no rmally without any treatment;②The rats in model group were infused with normal saline 5 mL/
kg per day;③The rats in the low dose guishen group were treated wi th guishen 2.1 g/kg per day;④The rats in middle dose guishen group were with g uishen 4.2 g/kg per day;⑤The rats in high dose guishen group were with guishen 8.4 g/kg per da;⑥The rats in oral solution group were treated with common self heal fruit spike(a kind of Chinese herb) oral solution 2.1 mL/kg per day. The m odels of
mammary gland hyperplasia were induced by benzoate estradiol intraperit oneal injection in SD female rats. Benovocylin was injected into the abdominal c avity to copy the models of rat mammary gland hyperplasia in all the groups exce pt the control group,and all the groups received corresponding treatment once pe r day for consecutive 4 weeks.At the end of 4 weeks,benovocylin was quitted to i nject,and corresponding medication continued for another two days. All rats were killed after the last medication to draw blood from hearts for the determinatio n of serum E2,P and PRL level with radioimmunoassay(RIA). Meanwhile,the tissue s ection of mammary glands was taken out and observed. RESULTS:All the 48 rats finished the result analysis. ①The level of serum E2 and PRL was significantly higher in the model group than in the control group<(6 8.28±57.61)
ng/L,(3.59±0.35) μg/L vs(16.06±2.36) ng/L,(2.87±0.40) μg/L>(P< 0.05 or 0.01);That in the low ,middle and high dose guishen group<(14.97±4. 92) ng/L,(2.95±0.53) μg/L,(11.56±4.00) ng/L,(2.75±0.61) μg/L,(15.50±3.43) ng/L,(2.39±0.67) μg/L respectively> was significantly lower than that in the m odel group(P< 0.05 or 0.01).②The number of mammary lobule and acinus was increa sed,lumen of catheter and alveolar cavity were obvioulsly enlarged in the model group. In the guishen capsule treatment groups,the number of mammary lobule and acinus was decreased to different extents,a large number of acinus was at break up and degradation,and lumen of catheter was contracted. CONCLUSION:Guishen capsule has obvious effect of decreasing the serum level of E2 and PRL. It can inhibit hyperplasia of mammary gland and regulate the endocr ine hormone balance.
