AIM:To compare the difference of the antidepressant effect between Jieyuwan an d its components on depressive mouse models. METHODS: The experiment was carried out in the pharmacological laboratory for neurology and psychology of the Department of Pharmacology,School of Basic Medic al Sciences,Peking University Health Science Center.Male ICR mice were randomly divided into 8 groups:control group(5 g/L carboxymethylcellulose sodium),fluoxet ine group (20 mg/kg),Jieyuwan small,middle and large dose groups (65.75,131.5 an d 263 mg/kg),Xiaoyaosan group (52.34 mg/kg),Yubaihe Fang group(14 mg/kg) and Gan mai Dazao Fang group(21.6 mg/kg).The antidepressant effects of Jieyuwan and its components were observed with two behavioral despair experiments of mice forced swimming test and mice tail suspension test.In order to identify whether the abo ve mentioned drugs had the role in central excitability,the automatic activitie s of the mice were determined to make sure the reliability of Jieyuwan and other antidepressants. RESULTS:The forced swimming test and tail suspension test showed that the immo bility time of rats was significantly shortened in the fluoxetine group,Jieyuwan middle and large dose groups,Yubaihe Fang group, Xiaoyaosan group and Ganmai Da zao Fang group(P< 0.
01).Compared with the fluoxetine group, there was on obvious difference in the Jieyuwan middle and large dose groups,Yubaihe Fang group,Xiao yaosan group and Ganmai Dazao Fang group(P >0.05).The immobility times in the Ji eyuwan middle and large dose groups were not obviously different from those in t he Yubaihe Fang group,Xiaoyaosan group and Ganmai Dazao Fang group(P >0.05).The automatic activities of the mice showed that their automatic activities were not notably affected by small,middle and large dose Jieyuwan,Yubaihe Fang, Xiaoyaos an and Ganmai Dazao Fang(P >0.05). CONCLUSION:Jieyuwan and its components can ameliorate the depressive behaviors of mice.It has the antidepressant effect but has no effect of central excitabil ity.The antidepressant effect of Jieyuwan is not different from those of its components.
