Objective To investigate the pharmacological effects of 4 kinds of effective fractions extracted from
Buyang Huanwu Decoction(BHD)
on the infiltration of polymorphonuclear leukocytes(PMNL)in the local cerebral focus induced by ischemia-reperfusion among rats.Methods The model of cerebral ischemia-reperfusion was prepared among rats with the method of nylon monofilament via the internal carotid artery to induce occlusion in the middle cerebral artery.Then,the 4 kinds of effective fraction were used to treat the modeling animals respectively and the activity determination of myeloperoxidase(MPO) in the cerebral tissue was taken to eveluate the degree of PMNL infiltration. Results MPO activity in cerebral tissues was significantly incerased after 46 hours of reperfusion following 2 hours of cerebral ischemia(P<0.05).When treated with the fractions of polysaccharide and alkaloid respectively to the modeling animals,MPO activities were all brought to a significartly decreasecl level in the local cerebral tissue of ischemia reperfusion(P<0.05).Conclusions Among the 4 kinds of effective fraction extracted from BHD,polysaccharide and alkaloid can decrease the activity of MPO in cerebral tissue.This means that PMNL infiltration will be inhibited in the cerebral tissue of reperfusion following
middle cerebral artery occlusion.It may be one of the mechanisms to treat cerebral ischemia reperfusion injury with BHD.
