Objective:In order to investigate the mutual actions among effecive components of medicinal Chinese herbs when making up
preparaˉtions.Methods:We use ACVas positive control of antiherpes and analyze the combinaiton effects between emodin and total saponins on HSVˉ2333
strain using Median Effect Principle with plaque reduction assays.Results:In Hepˉ2cell systems,the median effect doses(ED 50 )of ACV of inhiˉbition on the multiplication of HSVˉ2333Strain was10.85μm/ml.When emodin is combined with total saponins at the ratio of2(Dm)/1(Dm),the ED 50 s of direct virucidal activity,the inhibition effects on the infection and multiplication of HSVˉ2333strain are0.73μm/ml,1.04μm/ml and0.83μm/ml
respectively.When emodin is combined with total saponins at the ratio of1/Dm)/1(Dm),the corresponding ED 50 s are0.13μm/ml,0.45μm/ml and0.53μm/ml separately.When at the ratio of1(Dm)/1(Dm),the corresponding ED 50 s are0.17μm/ml,1.53μm/ml,0.99μm/ml respectively.The combination indexes of three combination ratios above are<1,and these results indicate that the two drugs have obvious antiherpes synergism.Conclusions:The TI of ACV to the inhibition of multuplication of HSVˉ2333strain is100.However,the TIs of2(Dm)emodin/1(Dm)total saponins of direct virucidal activity,the inhibition effects on the infection and multiplication of HSVˉ2are408.2,286.5,359.0respectively.The corresponding TIs of1(Dm)emodin/1(Dm)total saponins are1899.2,548.6,465.8separately.The correspondˉing TIs of1(Dm)emodin/2(Dm)total saponins are714.7,79.4,122.7respectively.Therefore,1(Dm)emodin/1(Dm)total saponins should be the ratio for first choice.
