Objective\ To study the characteristic of Ca\+\{2+\} channel mediated store\|operated Ca\+\{2+\} influx on rat vascular smooth
muscle.\ Methods\ Fura\|2 fluorescence technique was used to investigate the i change.\ Results\ (1)In smooth muscle cells of rat aorta 10 μmol/L cyclopiazonic acid(CPA) resulted biphasici change the transient peak phase of i and the sustained elevation(plateau) phase of i.\ (2) SK&F96365(5~40 μmol/L) produced inhibitory effect on CPA in a concentration\|dependent manner.\ The maximal inhibitory effect(50%±18%) of SK&F96365 was reached at a concentration of 20 μmol/L.\ (3)Inhibition rate of 1 μmol/L nifedipine on CPA\|induced increase ofi was 28%±7%, subsequent addition of 20 μmol/L SK&F96365 further strengthen the decrease effect.\ Total inhibition of both Nif and SK&F96365 was 60%±7%.\ Inhibition rate of 20 μmol/L SK&F96365 on CPA\|induced increase of i was 56%±11%, subsequent addition of 1 μmol/L Nif did not further produce any decrease effect.\ Conclusion\ That voltage\|dependent Ca\+\{2+\} channels and store\|operated Ca\+\{2+\} channels mediated store\|operated Ca\+\{2+\} influx induced by CPA, and there is an obviously inhibitory effect of SK&F96365 on voltage\|dependent Ca\+\{2+\} channels.\;
