\ Objective \ To investigate the influence of Kadsurenone (an antagonist of platelet activity factor
PAF) upon implantation
of mouse blastocyst.\ Methods \ Fiftytwo female healthy mice, mating
with male healthy mice of the same breed, in proportion as 2 to 1. The 1st day of pregnancy
was determined by the presence of vaginal plug. On the 2nd day of pregnancy, Kadsurenone in
different dosages were injected into the uterohorn of one side of mice, equal amount of
physiological saline were injected into the uterohorn of the other side. On the 8th day or the
10th day of pregnancy, the histological changes of uterus as well as fetus were observed after
the tissue being treated with
immunohistochemistry.\ Results \ In the experimental groups with
Kadsurenone 20L, the decidua became thinner , Laminin immunoreactivity reduced, while
fibronectin immunoreactivity increased, in association with retarded development and even
retrogressive degeneration of mouse fetuses compared with the control groups. In groups with
Kadsurenone40L, there's no
decidualization of uterus, the blastocyst was digsested. \
Conclusion \ By the mechanism of antiPAF , Kadsurenone impaired the decidualization of
uterus and influenced on the composition and distribution of extracellular matrix(e.g. Ln and
Fn), which might be one of the mechanisms of Kadsurenone against implantation. \
