OBJECTIVE: To establish a high performance liquid chromatography (HPLC) to determined the
concentration of
serum acetylleucomycin. This
method was evaluated with
microbiology method. METHODS: Blood samples were collected at various time after 10 male healthy volunteers oral administration of
acetylleucomycin granules. Serum concentration of drug were determined by HPLC and microbiology. RESULTS: Acetylleucomycin
standard solution and oral acetylleucomycin granules solution treated with active carbon were analyzed by HPLC. Its chomatography showed 8 components, A9 and A8 peaks were interfered with impurity in serum,other peaks were the same as that of standard reagent. A5 peak height of acetylleucomycin VS concentration of various standard drug solution gave the r=0. 9996( P < 0.0005 ) , and showed an ideal linear correlation in 0.026 - 3.000μg/ml concentrarion rang of drug. Average recovery ratio was 96. 43% .The practical lowest limit of detection was 0.026μg/ml. CVW was 4.75% and 5.80% at 0.5μg/ml and 1. 5μg/ml concentration of serum drug, resperctively. CVb was 6.42% and 6.70%. By microbiology method there was a linear corrlation beteen various concentration of acetylleucomycin standard solution and its diameter of inhibitory ring within 0.06 - 2.00μg/ml.Correlation coefficient was 0.9986.Practical lowest limit of detection was 0.06μg/ml. Coefficient of variationwere 5.07% - 8.8%. CONCLUSION: This results showed that HPLC was a good method of accuracy, sensitivity,precision for determined serum concentration of acetylleucomycin.
