pressure
was established according to motility of the isolated rat gastric muscle strip and water-sac perfusion manometry and ileum peristalsis of Guinea pigs. Intra-cavity pressure of isolated gastrointestinal canal of rat and guinea pig was observed.
Gastric intra-cavity pressure was markedly increased as added with various doses of acetylcholine (1.25, 2.5, 3.25, 5, 6.25 and 7.5 μg/mL respectively) in nutrient solution. The ileum intra-cavity pressure was also increased as added with various doses of histamine (0.125, 0.25, and 0.5 μg/mL respectively) . The effects were in a dose-effect manner and the differences were significant as compared with distilled water group ( P < 0.01 - 0.001). The rat gastric intra-cavity pressure declined in the perfusate added with 2.5 μg/mL acetylcholine and various doses of n-butyl alcohol extraction of Si Junzi Decoction (250, 500, 1 000, and 2 000 μg/mL respectively). 125 μg/mL n-butyl alcohol extraction of Si Junzi Decoction with 0.25μg/mL histamine had a slight excitation on the ileum of guinea pigs while large doese of n-butyl alcohol extraction of Si Junzi Decoction (250 -2 000 μg/mL) showed a inhibitory effect. The differences were significant as compared with distilled water group (P<0.05-0.001). < Conclusion> Acetylcholine in various doses can increase the isolated rat gastric intra-cavity pressure and histamine can increase the isolated guinea pig intra-cavity pressure. This indicates that the gastrointestinal motility was sensitive and this method is effective. n-butyl alcohol extraction of Si Junzi Decoction can inhibit the gastrointestinal motility.
