Objective To study the
lymphocytes proliferation stimulated by TFA GSA antigen in immune response of Guinea
pigs halothane hepatitis.Methods 30 male Hartley Guinea pigs were randomly divided into experimental group and control group.Both were sub divided again into 3 groups and each subgroup has 5 Guinea pigs.Experimental group:1% halothane and 40%O 2(equilibrated by N 2) were inhaled for 4 hours,re inhaled on the 42nd day or at the 84th day according to different subgroups,respectively once,twice and three times for the three subgroup.Control group:only inhaled 40% O 2.Lymphohocytes were separated on the 35th day in the first subgroup,63rd day in the second subgroup,105th day in the third subgroup,then were cultured respectively with different
concentration of TFA GSA antigen added. 3H TdR was added at 18 hours before the end of culture.The
lymphocytes proliferation stimulated by specific antigen during the process of immune response after halothane inhaled was analized by lymphocyte transformation test(LTT).Result Specific antigen induced lymphocytes proliferation of Guinea pig hahaled halothane.The minimum valuable concentration decreased with the increase of inhaling halothane times.Conclusion Specific antigen promoted lymphocytes proliferation,the minimum positive concentration and the most suitable concentration were related to the times of halothane inhalation.Excessive antigen could lead to immune tolerance. <
