Objective To investigate the protective effects of
Jiedutongluobaoshen Capsules (JC) against
renal lesion in
diabetic rats.Method The experimental animals were divided into the normal control group, diabetic control group, positive control group and experimental group. The levels of blood sugar, urea nitrogen, blood creatinine, and
angiotensin Ⅱ (Ang Ⅱ) were measured, and the excretion rate (UAER) of urinary albumin and the index of renal hypertrophy were detected in each group at the 12 th week after the experiment. At the same time, RT-PCR was used to detect the mRNA
expression of renal cortex
connective tissue growth factor (CTGF), and immunohistochemical techniques were used to detect the expressions of renal fibronectin (FN) and type IV collagen (COL Ⅳ).Results In the JC-treated group, the blood sugar level was markedly lowered(P<0.01), UAER was markedly reduced (P<0.05), the levels of urea nitrogen and blood creatinine were decreased, and the renal hypertrophy index was improved(P<0.01), as compared with those in the diabetic control group. The RT-PCR detection showed that the mRNA expression of renal cortex
CTGF was increased and the sediment of glomerular FN and Col Ⅳ was raised in the diabetic control group, while in the JC-treated group, the expressions of CTGF mRNA, FN and Col IV were lower than those in the diabetic control group (P<0.05). Conclusion The mechanism of the protective effects of JC against renal lesion may be decreasing the level of Ang Ⅱ in the renal tissue, downregulating the expression of CTCF, and reducing the sediment of extracelluar matrix.
