Objective To investigate the effects of
glutathione(GSH) on
cisplatin-induced toxicity to renal tubular epithelial cells(RTEC)isolated
from different age rats.Methods RTEC were isolated from 3-week-old,2-month-old or 5-month-old rats and inoculated into 96 well plates,after 24 hours' preincubation,a series of concentrations of
cisplatin,in the presence and absence of L-cysteine or DL-buthionine-(S,R)-sulfoximine(BSO),were added,and after 24 hours' coincubation,MTT assays were performed to test cell viability.Results The concentrations of cisplatin that inhibited 50% cell growth(IC_ 50 ) of RTEC from 5-month-old,2-month-old or 3-week old rats were 243\^42,178\^16 and 159\^06 μmol/L,respectively,BSO made IC_ 50 of cisplatin lower down to 1\^62,1\^30 and 1\^47 μmol/L,respectively,while L-cysteine made IC_ 50 of Cisplatin increase up to more than 5 000 μmol/L. Conclusion Cisplatin is dramatically toxic to RTEC from different age rats;BSO can enhance Cisplatin-induced cytotoxicity,while L-cysteine can protect it,it is indirectly proved that GSH can protect cisplatin-induced toxicity to RTEC from rats.
