AIM To investigate the protective effects of CAT or F IS from Rosa bracteata Wendl on the iron-loaded rat hepatocytes. METHODS
Iron over-loaded model was induced by addition of 50 μmol ·L -1 iron citrate to the rat hepatocytes. At the meantime or 24 h after iron treatment, CAT, FIS or Dereroxamine was added to the culture medium . At 24 h after the drug treatment, the activities of SOD, LDH, ALT and AST of the in rat hepatocyte cultures were measured. At 48 h after the drug treatment, the survival rate of Hepatocytes was measured. RESULTS CAT and FIS h ave less effect on the rat hepatocytes treated with the iron and drug together, while the group of CAT 5×10 -5 、5×10 -6 、5×10 -7 mol·L -1 ,FIS 5×10 -5 , 5×10 -6 , 5×10 -7 mol·L -1 DFO treated w ith iron first could raise the content of SOD, restrain the release of AST and A LT, lower the level of LDH and the content of ALB on those treated with the iron first. The survival rate of hepatocytes also increased. CONCLUSION CAT and FIS have a good protection against the iron toxicity in rat hepatocyte c ultures.
