Objective: To establish a HPLC method for determination of 17997 in plasma and to study its pharmaco-kinelics in rabbits
by iv
injection. Methods: Doses of 4,2 and 1 mg · kg-1 were administrated to three groups of rabbits by iv injection. Blood samples were collected during 6 hours in accordance with the experimental design. The drug levels in plasma were measured by HPLC assay. The time course of mean plasma concentration was simulated by NDST21 program software. Results:The concentrations in plasma at different time have been measured and better fitted the two - compartment model with first kinetics process. The main
pharmacokinetic parameters of the three doses of 4,2 and 1 mg·kg-1 were followed : T1/2β were 46.62,48.20 and 42. 16 min; C0 were 4. 15,2. 14 and 1.58 μg · mL-1 ;CL were 41. 2 ,49. 3 and 47. 6 mL · min-1 · kg-1 ; AUC were 97. 22,40. 54 and 21. 00 min · μg · mL Respectively. Conclusion: We have successfully used the developed analytical method for 17997 to study its pharmacokinetical properties of rabbits after a rapid iv injection. The method is found to be sensitive, simple and rapid , suitable for application in preclinical and clinical pharmacokinetic studies. The distribution and metabolism of 17997 is fast.
