AIM To develop a HPLC method for simultaneous determination of phenobarbital(PB), phenytoin(PT), and carbamazepine(CBZ) in human serum. METHODS The HPLC assay was established by C 18 column with methanol water(55∶45) as the mobile phase, barbital(BB) as internal standard and the detection wavelength at 220 nm. The flow rate was 1 0 ml·min -1 . The temperature of the column was 30℃. RESULTS The linear range of PB was over 1 0~80 mg·L -1 with a correlation coefficient of 0 999 9 . The linear range of PT was over 1 0~50 mg·L -1 with a correlation coefficient of 0 999 6 . The linear range of CBZ was over 0 5~20 mg·L -1 with a correlation coefficient of 0 999 5 . The limit of detection was 0 25,0 25,and 0 1 mg·L -1 , respectively. The average recovery of the three drugs were 95 75%, 96 54%, 92 13%, respectively. The intra day RSD(%) was less than 6 29% and the inter day RSD(%) less than 6 64%( n =5). CONCLUSION This method was found to be simple, quick, and sensitive. It can be used for monitoring therapeutic drugs and studying pharmacokinetics especially for the epileptic patients.
