Objective:To establish
capillary electrophoretic(CE) method for the
separation and determination of
cefaclor in human
urine.Methods:Urine
samples were injected directly into the capillary after multi fold dilution.The electrophoretic separation was achieved with 50 mmol/L phosphate 12.5 mmol/L borate (pH 8.08 ) running buffer and a constant voltage of 20 kV was
applied to the 48 cm×50 μm uncoated capillary(Supelco, USA). The analytes were introduced into capillary by pressure(8.274×10 4 Pa·s) and determined with on column monitoring at 265 nm.Results:An ideal separation for cefaclor and internal
standard cefradine in human urine was completed within 6.5 min by CE. The analytical results of cefaclor urine samples demonstrated a good linear relationship within the range of 2.5 250.0 μg/ml. The absolute recoveries were more than 95.5% and the relative standard deviations of inter day and intra day were less than 8.0%.Conclusion:The CE method is accurate, simple, rapid and has been applied successfully in dynamic analysis of urine samples after a single oral dose of 500 mg cefaclor capsule in 6 male healthy volunteers.
