The major pathological change in hypertensive kidney is glomerular sclerosis with unknown mechanism. With the development
of molecular biology, it has been found that polypeptide growth factors, such as bFGF and ET might play an important role in the pathogenesis of glomerular disease. By measuring the
expression of bFGF in kidney and the ET level both in plasma and in renal tissue, we investigated the mechanism of renal damage in SHR . There were seven 16 month old SHRs in experimental group and seven WKY of same age in the control group. Serum BUN and Cr as well as 24 hour CCr were measured. UAE and ET level both in plasma and kidney were
detected by RIA. Renal sections were stained by HE and PAS. Expression of collagen Ⅰ,Ⅳ and bFGF in renal tissue were detected immunohistochemically. The bFGF mRNA expression was detected by in situ hybridization. There were marked functional and pathological changes in SHR with glomerular sclerosis. ET in plasma and renal tissue in SHR was increased significantly, the espression of bFGF in kidney was greatly enhanced. ET and bFGF might be involved in the renal damage in SHR.
