Objective:To study the cytotoxicity of soman during hypoxia on PC12 cells and the mechanism of increasing toxicity of organophosphate
compounds under the condition of hypoxia.Methods:The cytotoxicity was measured by MTT spectrophotometric method,the concentration of cAMP and 〔Ca 2+ 〕i were detected with RIA and fluorescent probe (Fura 2/AM) and protein content was assayed by Folin phenol method.Results:Both hypoxia andsoman intoxication with hypoxia remarkably reduced the survival rate of PC12 cells,but in soman intoxication alone no obvious change of survival rate was found;the concentration of cAMP and〔Ca 2+ 〕i in PC12 cells were remarkably increased in hypoxia, soman intoxication and soman intoxication with hypoxia groups. The changes in soman intoxication with hypoxia group were more significant(p<0.01).Conclusion:The increase of signal transduction and the exacerbation of neurotoxicity on PC12 cells induced by hypoxia may play an important role in the increase of toxicity of organophosphate compounds during hypoxia.
