OBJECTIVE To study the preparation and in vivo disposition through skin absorption of liposomal gel including psoralen
and
dexamethasone acetate in order to develop a new therapeutic preparation for vitiligo.METHODS To screen the composition and preparing method using homogeneous design. Its outward appearance and size distribution were detected by scanning electron microscope (SEM).Molecular sieve and HPLC were used to study the encapsulated rate of the liposomal gel .We also observed its stability in vitro preliminarily. Finally we tested its skin irritation and in vivo disposition through skin absorption. RESULTS The liposomal gel had promising characters of fine looking appearance, shallow size distribution, high encapsulated rate and good stability in vitro . Compared with the control, it had significant protective action for rabbits' skin.It produced higher accumulation of psoralen and
Dexamethasone acetate in skin(4.2 fold, P < 0.01 ) and low drug concentration in the internal organs of guinea pigs ( P <0.05).CONCLUSION The results strongly suggested that the liposomal gel including psoralen and dexamethasone acetate was valuable to be further developed as a new therapeutic preparation for vitiligo with high effect and low toxicity.
