AIM: To establish determination method of the concentration of
cefazolin in serum and articular cavity fluid,and to observe
the distribution of the cefazolin in the patients of septic knee or osteal arthritis with contraction for providing bases to clinical rational drug uses and to increase treatment effect. METHODS: Cefazolin concentration in serum and
articular cavity fluid after 0, 0 5, 1, 1 5, 2, 2 5, 3, 5, 7 and 24 h was determined with reversed phase
high performance liquid chromatography (RP HPLC). The dosage of cefazolin was 70 mg/kg and add to 250 ml of normal saline and to intravenous drip for 30 min. RESULTS: The linear relation between cefazolin concentration in the range of 2-100, 100-500 μg/ml and the rate of peak areas were all right. The individual difference of the distributive concentration in the serum and the articular cavity fluid was major. The relativity between the cefazolin concentration and the rate of peak areas was not prominent. The time of coming to the peak of articular cavity fluid laged behind the serum more than 1 5 h. CONCLUSION: Determination of the cefazolin concentration of the body fluid is simply and reliable with this method. The method can satisfy the requirement of the drugs monitoring of the clinical treat.
