QBJECTIVE To establish reverse high-performance liquid chromatography in the determination of glycyrrhizin in the plasma
of rats with spleen deficiency and investigate into the pharmacokinetics of glycyrrhizin in rats with spleen deficiency. METHOD Internal stan- dard: diphenyl; mobile phase: methanol-water-acetic acid (80: 19: 1, v/v); stationary phase: ODS-3 chromatographic column (150 mm x 4. 6 mm, 5 μm); detection wave length: 250 nm; flow rate: 1 .0 mL/min. Muscular injection of reserpine (0.3 mg/kg) was given to rats daily for 14 days before splgeen deficiency - were made. Glycyrrhizin was given to the rats after 14 hours of empty stomach and plasma was obtained after 5, 10,20,30,90,120 and 180 minutes. Blood concentration was then determined and time data obtained, which were fitted with 3P87 software.RESULT Minimum limit of detection of this method: 2 ng.; CV% < 10%; recovery rate: 96.20% ~ 103 .21 %, with satisfactory linear relation within the range of 40. 8 ~ 1467. 8 μg/mL; PK: open bicameral model. CONCLUSION This method is simple, rapid, sensitive and accurate, and can be used to determine rat plasma glycyrrhizin.
