Objective To select an accurate method for determining the protein contents in
earthworm extract. Methods Protein contents
in
earthworm extract and in bovine serum albumin (BSA) solution before and after enzymolysis were determined using Lowry's modified assay and Bradford's dye-binding assay respectively for comparing the difference between both. Results The protein contents in
Earthworm extract by Lowry's modified assay were higher than that by Bradford's dye-binding assay. Before hydrolysis,the protein contents in BSA solution by the two methods were similar. After hydrolysis, the contents determined by Lowry's modified assay were slight higher. On the contrary, the contents determined by Bradford's dye-binding assay were much lower. Conclusion Bradford's dye-binding assay cannot effect the contents of peptide, while the contents of BSA after hydrolysis are much lower.It indicats that Lowry's modified assay is sensitive to proteins and peptides in earthworm extract and more suitable for determining the protein contents in earthworm extract.
