Objective To study the effect of Ganyanping on transforming growth factor-β_1(TGF-β_1) and hepatocyte growth factor(HGF) of rabbit model of schistosmal liver fibrosis.Methods The rabbits were separated randomly into three groups: normal control group(N),model group(M) and Ganyanping-treated group(E) those in.The rabbits in groups M and E were infected with cercariae of Schistosomiasis japonicum(180 cercariae per rabbit) and those in group E treated with Ganyanping for 6 weeks after the model was made successfully.The effect of Ganyanping was explored by the expressions and sites of HGF and TGF-β_1 in liver tissues by immunohistochemical staining.The protein levels of HGF and TGF-β_1 were analyzed by Western blotting.RT-PCR was used to analysis the mRNA level of HGF and TGF-β_1.Results Compared with the group M,the espression of HGF was increased,while the expression of TGF-β_1 decreased in the liver in the group E.The difference was significant(P<0.05).Conclusion Ganyanping could inhibit the expression of TGF-β_1 and enhance the expression of HGF in the liver of rabbit with schistosomal liver fibrosis,which may be one of the antifibrotic mechanism of Ganyanping.
