Objective To investigate the effects of Dexamethasone on
degradation of
corneal fibroblasts induced by Neutrophil.Methods The corneal fibroblasts -
collagen gels were incubated with Neutrophil or Neutrophil-conditionde medium(CM) and different concentrations of Dexamethasone or in MEM culture medium separately 48 h later,to evaluate the collagen degradation level,and the amount of hydroxyprline(HYP) were measured.The release of MMPs into the culture supernatant was assessed by Immunoblot Analysis.Results In the presence of plasminogen,the amount of HYP was increased with the Neutrophils or CM.There were
significant differences(P<(0.01)).Dexamethasone inhibited the Neutrophil-induced degradation of collagen by corneal
Fibroblasts in a dose-dependent manner.There were significant differences(P<(0.01)).The Neutrophils-CM stimulated the synthesis of matrix metalloproteinase(MMP)-1 and MMP-3 by corneal fibroblasts.Conclusion Dexamethasone inhibits collagen degradation by corneal fibroblasts.Neutrophils increased collagen degradation by corneal fibroblasts.
