Objective: To investigate the role of
glycyrrhizin on
proliferation, activation, synthesis of extracellular matrix in rat
hepatic stellate cell(HSC). Methods:The mice HSC were isolated and cultured in vitro. Samples were extracted from control and
glycyrrhizin groups. 1 to 1000μmol/1 glycyrrhizin was incubated with HSC; The inhibitory rate of
proliferation was measured by MTT method, the viability of cells was tested by evaluation of supernant LDH and Trypan blue dye exclusion. α-SMA,collagen I,Ⅲ were analyzed with Western blot. Results: MTT method showed an obvious inhibition of HSC proliferation after 100 to 1000μmol/L glycyrrhizin treatment. Different concentrations of glycyrrhizin did not affect the vitality on HSC by LDH measurement and Trypan Blue dye exclusion method. 1 to 1000μmol/l Glycyrrhizin could inhibited protein expression of α-SMA, collagen I,Ⅲ gradually. Conclusion: Intervention of HSC proliferation, activation and decreasing the synthesis of collage may contribute to the anti-fibrosis molecular mechanism of glycyrrhizia
